* 投稿摘要: By immunohistochemistry, a mammalian ste20-like kinase 3 (MST3), was mostly stained in the medulla of the kidney of B6 mice and Wistar-Kyoto (WKY) rats. Furthermore, MST3 levels were markedly reduced in the principle cell in the medulla of spontaneously hypertensive rats (SHR) compared with WKY controls. The degree of the reduction was augmented with prominent apical site of the principle cells of the collecting duct. The MST3 level was also measured before (age 3-4 weeks) and after (age 12-14 weeks) the development of hypertension and in SHR and age-matched WKY controls by western. 12-14-week-old SHR also had significantly reduced MST3 in outer and inner medulla compared with those of WKY controls. The difference in MST3 levels already existed between prehypertensive SHR and WKY rats. In vivo, the urinary sodium and water excretion from the urine was higher in constitutive MST3 transgenic mice on high salt diet compared to WT-MST3 mice on same diet. It raises the possibility that decreased inner medullary collecting duct production of MST3 could contribute to inappropriate sodium and water retention. Finally, 3B5 cells expressing WT-MST3 persists to NaCl but not urea induced perturbing effects of hyperosmolality at osmotic concentrations of 1,200 mOsm/Kg H2O. The present studies show for the first time that MST3 decreasing in inner medullary collecting duct is an intermediate kinase to inhibit urinary sodium retention in hypertension. The identification of MST3 might uncover a brand new gene that could prosper into a novel diagnostic and/or therapeutic target for hypertension. p.s. I regret that there is no data from human beings. We may wait till the chance comes and perform the experiment in the future.
