* 投稿摘要: Mitochondrial dysfunction has been suggested to participate in the pathogenesis of type 2 diabetes mellitus (T2DM). Among the mitochondria-related studies, effects of PTEN-induced kinase 1 (PINK1) on the mitochondrial function is extensively studied. PINK1 is a mitochondrial kinase with the function of protecting cells from stress-induced mitochondrial dysfunction. Several studies indicate that knockdown of PINK1 decreases glucose uptake and expression of fatty acid-binding protein in vitro. Accordingly, we hypothesize that PINK1 may participate in glucose and lipid metabolism as well as enhance insulin action, and therefore, involve in diabetic pathogenesis. To elucidate the roles of PINK1 in energy homeostasis, the present study aimed at addressing the putative involvement of PINK1 in lipid metabolism by examining PINK1 protein expression patterns during adipocyte differentiation (adipogenesis). Our results indicated that full length PINK1 was processed to smaller protein fragments during adipogenesis, while inhibition of adipogenesis abolished this proteolytic processing. In addition, the filamentous mitochondria in pre-adipocytes turned into punctuated morphology which was accompanied with the decrease of mitochondrial DNA copy number during the adipogenic process. Taken together, the proteolytic processing of PINK1 occurs simultaneously with alteration of mitochondrial morphology and copy number during adipogenesis. The above results suggest that PINK1 may play an important role in adipogenesis and energy metabolism through regulating mitochondrial morphology and function.
