* 投稿摘要: Type 2 diabetes mellitus (T2DM) is associated with distinctive abnormalities of insulin and glucose metabolism, often designated as insulin resistance. Our previous results show that body weight gain, fat mass and lipid droplets in adipose tissues of interleukin-4 (IL-4) treated mice are significantly reduced while circulatory adipokines and free fatty acids (FFAs) are increased. The above results suggest that IL-4 may inhibit lipid accumulation. Accordingly, it is tempting to further explore the regulatory mechanism of IL-4 to lipid metabolism. The major aim of the current study is to identify the putative regulation of IL-4 to expression and activity of hormone-sensitive lipase (HSL), the rate-limiting enzyme in the degradation of triacylglycerol to glycerol and FFAs. Our data showed that IL-4 not only promoted HSL activity by increasing HSL phosphorylation at Ser563 residue but also translocation of HSL form cytoplasm to the surface of lipid droplets for its getting access to the lipid core. The enhancement of HSL phosphorylation and translocation by IL-4 was attenuated by H89, a potent protein kinase A inhibitor. The above results suggest that IL-4 plays a pro-lipolytic role in lipid metabolism by boosting HSL activity through protein kinase A pathway.
