* 投稿摘要: Tuberculosis (TB) recently has re-emerged as a major public health threat worldwide. However, there is strong evidence that host genetic factors influence individual susceptibility to tuberculosis. The association between polymorphisms of innate immunity receptors cluster of CD14, mannose-binding lectin (MBL), Toll-like receptor (TLR)-7 and TLR8 with susceptibility in 78 patients with active TB and 243 healthy controls were investigated. Genotyping for CD14-159, TLR7 IVS2-151, TLR8-129 were performed by polymerase chain reaction-restriction fragment length polymorphism methods, MBL polymorphisms were analyzed by TaqMan 5´ nuclease assay in Real-Time PCR System. In this study, C allele in TLR8 promoter -129 was demonstrated to be significantly higher in patients with TB infection compared to controls (22.1% vs. 9.5%; OR:2.71; p=0.005). The B allele in MBL codon 54 conferred a protective role against TB in our study population with a significant difference (OR = 0.58; p = 0.05). But subjects with H allele in MBL-550 had a higher risk to susceptibility to TB (OR = 1.84; p = 0.0006). However, no significantly association between CD14 -260 G>C and TLR7 IVS2-151 G>A polymorphisms with TB infection was found. In conclusion, our results provide an evidence of the association between MBL and TLR8 gene polymorphism with susceptibility to TB infection, but not the SNPs of CD14 and TLR7 in our population.
