* 投稿摘要： Overexpression of the efflux pump AdeABC is associated with tigecycline resistance of multi-drug resistant Acinectobacter baumannii (MDRAB). The pump is regulated by a two-component regulatory system, sensor AdeS and regulator AdeR proteins. However, the detailed mechanism of the AdeR protein to enhance the expression of adeABC operon is not well defined. A wild type A. baumannii (AB260) and a clinical isolate (AB293) of TGC-resistant MDRAB with two point mutations (Met197Ile, Ser200Cys) on its AdeR protein were included in this study. AdeR proteins were applied to investigate the different binding characteristics between the promoter and the intercistronic spacer DNA fragment between adeRS and adeABC genes. By electrophoresis mobility shift assay, neither wild type nor mutant AdeR protein bound to the adeABC promoter DNA fragment. However, wild type AdeR protein strongly bound to the intercistronic spacer DNA fragment and the mutant AdeR protein demonstrated about 100 times lower binding activity to the same DNA fragment. A direct repeat motif was observed in the intercistronic spacer DNA and considered to be the AdeR binding site. Two residue mutations on AdeR protein may liberate the intercistronic spacer from binding. The free intercistronic spacer may support the successful read-through of adeABC operon by the cotranscriptional translation machinery in A. baumannii.
