* 投稿摘要: Melanoma is the most malignant cancer in skin cancer and it’s a fatal cancer type that account for 80% death in skin cancer in the USA, UK and other EU country. At early stage of melanoma, surgery is mainly treatment for patients and its’ 5 year survival rate up to 90%, however, it declined to below 60% in postoperative with chemotherapy or radiotherapy. In traditional medicine, native plants can be used as herbal medicine for the treatment or prevention of disease, but also as a dietary medicine or food daily. Previous studies, HeJuCi is used for treatment of acne, hernia, relieve pain, diuretic and stomach, can promote liver detoxification and strengthening liver function, but also can be used to eliminate the congestion. For aspects of joint and muscle, HeJuCi can relieve gout and rheumatism. In recent years, studies indicate that this plant extracts inhibited colorectal cancer and leukemia cells growth, but the detailed mechanisms have not been verified. The aim of this project is to research the anti-cancer effects and mechanisms of HeJuCi extract on melanoma. At the first, the MTT assay was performing to test the cytotoxicity of HeJuCi extract in melanoma and normal cells and the results indicated that HeJuCi extract inhibited B16/F10 cell growth and less toxicity to normal cells comparison of 5-FU. Next, HeJuCi extract combined with 5-FU that enhanced the cytotoxicity to B16/F10 cell had synergistic effect with CI value less than 1. To get insight to explore the anti-cancer mechanisms of HeJuCi extract on B16/F10 cells, the results demonstrated that HeJuCi extract induced cell cycle arrest at G0/G1 phase and also increased the sub-G1 phase cell population in a time and dose dependent manner that was analyzed by flow cytometry after HeJuCi extract treatment. To confirm that HeJuCi extract might altered the expression of several cell cycle related proteins (Rb, Cdk2, Cdk4, CyclinA, CyclinB1, CyclinD1) and cell cycle regulators (p21, p53 and p-p53), and the results showed that the protein expression of cell cycle related proteins reduced and cell cycle regulators were activated. On the other hand, to examine whether HeJuCi extract led to melanoma cell death or not, the results demonstrated that HeJuCi extract caused B16/F10 cell death, forming the apoptotic bodies, DNA fragments, DNA condensation and anoikis in cell morphology that performing by TUNEL assay and HeJuCi extract could activate the extrinsic and intrinsic apoptosis pathway to trigger cell death. In addition, HeJuCi extract also reduced the angiogenesis and metastasis protein expression. In conclusion, HeJuCi extract could inhibit melanoma cell growth by induced cell cycle arrest, activated cell apoptosis and suppresses angiogenesis and metastasis protein expression. Therefore, HeJuCi extract might provide a new therapeutic agent for melanoma cancer treatment.
