* 投稿摘要: Curcumin (Cur) is a commonly used coloring agent and spice in food. Previously, we reported curcumin blocks influenza A virus (IAV) activity via multiple mechanisms. By comparing the effect of curcumin analogues on IAV infection, monoacetylcurcumin (MAC) harbors a similar anti-IAV strength to curcumin. However, unlike curcumin, MAC did not inhibit viral HA activity, indicating that MAC possibly blocks IAV via a different mechanism. Hence, in the study herein, effect of these two compounds on several aspects related to IAV infection was compared. First of all, results of liposome assay and MUNANA test showed that as with curcumin, MAC attacked membrane integrity and mildly inhibited the activity of viral envelop protein NA. Accumulated evidence demonstrated that curcumin and its analogues inhibit several cellular signal transduction pathways, such as Akt and NF-kappaB signaling that are activated by IAV and also required for IAV replication. Treatment of curcumin analogues after virus entry indicated that curcumin and MAC inhibited phosphorylation (activation) of Akt and NF-kappaB to a similar extent. Finally, analysis two early events of infection indicated both MAC and curcumin strongly inhibited membrane fusion activity and viral RNA replication. Taken together, as a strong inhibitor of IAV, MAC shares similar effect to curcumin, but it does not inhibit HA activity.
