* 投稿摘要: Oral cancer is one of the ten most common cancers in the world. Furthermore, the incidence of oral cancer is higher in Taiwan due to the habits of tobacco, alcohol and areca use. The 5-year survival rate of oral cancer is 63%, but it declined to 37% with distant metastasis stage. Besides, about 18% of patients have distant metastasis at the time of diagnosis. Therefore, it is urgent required to development a new anti-cancer agent. In traditional medicine, PuJiCu can be used on anti-acne, aromatherapy, bladder infection, cellulite, and detoxification and so on. Thus, the purpose of this project is to investigate the anti-cancer effects and mechanisms of PuJiCu extract in oral cancer. In previous screening experiments, the results showed that both PuJiCu extract and 5-FU could inhibit oral cancer cells growth in a dose and time dependent manner, however, the cytotoxicity of 5-FU is higher than PuJiCu extract treatment in normal cells. Additionally, PuJiCu extract combined with 5-FU had synergistic effect, with the combination index (CI) <1, and the cytotoxicity of combinatorial treatment was greater than 5-FU along. Next, we focused on the anti-cancer effects of PuJiCu extract in vitro. At first, treatment with PuJiCu extract, the cell cycle distribution was analyzed by flow cytometry and these results showed that PuJiCu extract induced G0/G1 phase arrest and increased sub-G1 phase cell population. To determine whether PuJiCu extract induced cell apoptosis in oral cancer cells, TUNEL assay was performing to observe cell death morphology. The results demonstrated that oral cancer cells were formed apoptosis morphology including apoptotic bodies, DNA fragmentation, DNA condensation and anoikis after PuJiCu extract treatment indicated that PuJiCu extract could induce cell apoptosis. PuJiCu extract treatment also led to altered the expression of several cell cycle related proteins (p-Rb, Rb, Cdk2, Cdk4, CyclinA, CyclinB1, CyclinD1) and cell cycle regulators (p21, p53 and p-p53). To gain insight into the mechanisms of apoptosis, the results indicated that both extrinsic pathway (Fas, FasL and caspase 8) and intrinsic pathway (Bax and caspase 9) were activated, and the expression of apoptosis related protein (pro-caspase 3, 8 and 9) were reduced after PuJiCu extract treatment. Furthermore, PuJiCu extract treatment could reduce the expression of angiogenesis and metastasis related protein in oral cancer cells. In conclusion, PuJiCu extract could induce cell cycle arrest at G0/G1 phase and through extrinsic and intrinsic pathway to trigger cell apoptosis. The combination of PuJiCu extract with 5-FU also had synergistic effects in oral cancer cells. Besides, PuJiCu extract could reduce angiogenesis and metastasis related proteins expression in oral cancer cells. Therefore, PuJiCu extract may be a potential chemotherapy agent in oral cancer therapy.
