* 投稿摘要：	Hepatocellular carcinoma (HCC) is the fifth in most common cancer and the second leading cause of cancer-related deaths worldwide. Additionally, the poor prognosis of HCC is related to delay diagnosis at late stage and chemotherapy drug resistance caused average lifespan below 10.9 months. Therefore, it is urgent require for an anti-cancer drug of HCC therapy. In traditional medicine, FrJNCM extract had been used to improve detoxification, eczema, nerve pain, and so on. Recent studies show that FrJNCM extract have anti-cancer activities and can induce cell apoptosis on breast cancer cells. However, the detail mechanisms of FrJNCM extract on HCC is unclear. Consequently, the purpose of this study is to investigate the anti-cancer effects and mechanisms of FrJNCM extract on HCC in vitro and in vivo. The results showed that FrJNCM extract could inhibit HCC cells growth and had lower cytotocixity in normal cells than tumor cells. In addition, the combination index (CI) value was 0.23 indicating that FrJNCM extract had synergistic effect that combined with VP-16 clinical drug. Furthermore, FrJNCM extract could induce cell cycle arrest at G0/G1 phase and increase cell number of sub-G1 phase with time course and dosage dependent manner, suggesting that FrJNCM extract could inhibit HCC cell proliferation. After FrJNCM extract treated HCC cells, results showed TUNEL positive and apoptosis morphology, such as anoikis, chromosome condensation, DNA fragmentation, and apoptotic bodies. By immunocytochemistry (ICC) staining, the tumor suppressor proteins (P53, P-P53) and cyclin-depenent kinase inhibitor (P21) were increased, and cell cycle associated proteins (Cyclin A, Cyclin B1, Cyclin D1, CDK2, CDK4) were decreased. Moreover, apoptosis associated molecules were activated, including extrinsic pathway (FAS, FASL, cleavage caspase-8), intrinsic pathway (AIF, Bax, cleavage caspase-9) and cleavage caspase-3. In animal study, nude mice bearing HepG2 were treated with vehicle and FrJNCM extract. The result revealed that FrJNCM extract could suppress tumor growth, prolong animal survival time and had low or no physiological and pathological toxicity. In conclusion, FrJNCM extract had strongly anti-cancer effect in vitro and in vivo. Therefore, FrJNCM extract has highly potential to develop as a new anti-cancer agent in HCC therapy.
