* 投稿摘要: Over the last few decades, multidrug-resistant (MDR) A. baumannii has gradually emerged as an important nosocomial pathogen worldwide. Carbapenems, primarily, imipenem and meropenem, have been used to treat MDR A. baumannii infections. Recently, there are increasing reports of carbapenem-resistant A. baumannii being isolated. However, little is known about the effects that acquisition of imipenem resisitance on the virulence and fitness of A. baumannii. In order to assess the fitness and virulence impacts of imipenem resistance, an imipenem selected mutants was generated from imipenem susceptible strain ATCC 17978 by multistep selection resistance. A. baumannii ATCC 17978 and its induced imipenem-resistant derivatives, were analyzed for in vitro growth rate, detergent-serum resistance, biofilm formation and live-dead fluorescence microscopy assay. The in vitro growth rate of the imipenem-resistant mutant was similar to wild type strain. Detergent-serum resistance assays showed that the induced imipenem-resistant A. baumannii with increased resistance to detergent and serum. Biofilm formation assay showed that the imipenem-resistant mutant produced significantly more biofilm than the wild type strain. Furthermore, the live-dead assay result showed the induced imipenem-resistant A. baumannii exhibited higher toxicity to A549 cells than wild type. Based on these results, we show that there is significant impact on fitness and virulence after acquisition of imipenem resistance. We hope this research would be the basis of eliminating drug-resistant strains, and promote the further development of new antimicrobial agents in the future.
