* 投稿摘要: Helicobacter pylori are microaerophilic, spiral-shaped Gram negative bacteria, which infect the human stomach in approximately half the population over a lifetime. Persistent infection with H. pylori that colonizes the human gastric or duodenal epithelium, causes gastric atrophy, gastric ulcer, duodenal ulcer, and acute/chronic gastritis, and possibly leads to severe gastroduodenal diseases such as gastric cancer or lymphoma. Numerous virulence determinants of H. pylori have been identified. Among them, type 1 capsular polysaccharide biosynthesis protein J (CapJ) is the most important which can promote H. pylori to take up cellular cholesterol. Infection of H. pylori induces autophagy that may facilitate bacterial survival in the cells. However, whether CapJ is involved in H. pylori-induced autophagy and survival in macrophages have not been investigated. In this study we found that wild type (WT) H. pylori possessed CapJ which induced autophagic pathway, attenuated lysosome fused with autophagosome, and led to more bacterial survival in macrophages when compared to capJ knockout mutant (∆CapJ). Depletion of cholesterol inhibited autophagosomes formation by H. pylori. Knockdown autophagy-associated molecule, Atg12, impaired autophagy followed by reduction of H. pylori multiply in macrophages. These results demonstrate that cholesterol glycosylation is involved in H. pylori survived intracellularly in macrophages and that is mediated in a cholesterol-dependent manner.
