* 投稿摘要: The emergence and spread of carbapenem-resistant Acinetobacter baumannii poses a challenge for optimizing antibiotic therapies and preventing outbreaks. Traditional phenotypic assays such as the Modified Hodge Test (MHT) or polymerase chain reaction-based detection of the carbapenemase genes are time-consuming and complicated. Therefore, new approaches for the efficient detection of carbapenemase-producing A. baumannii are urgently required. In this study, we use nanodiamond as solid extraction tool to extract supernatant proteins of test strains of A. baumannii and further analyze with matrix-assisted laser desorption/ionization mass spectrometry (MALDI-TOF MS). We acquire simple mass spectra of carbapenemase-producing A. baumannii and discriminate the non-carbapenemase-producing A. baumannii in less than 2 hours. A total of 32 A. baumannii strains were used in this study, including 25 clinical isolates with 15 carbapenem-resistant strains and 10 carbapenem-susceptible strains, 6 ATCC strains and 1 selected mutants. The results of the nanodiamonds coupled with MALDI-TOF MS assay showed the same sensitivity compared to the results of the MHT. Using this method, the carbapenemase is directly detected and would not be affected by the genotype diversity, unlike molecular detection, thus making it more efficient. Our results demonstrate the ability of the former method to routinely detect carbapenemase-producing A. baumannii.
