* 投稿摘要: Backgrounds: EV71 infections could cause neurological diseases in young children. Type I interferon (IFN) signaling is important for innate antiviral responses. However, EV71 have evolved measures to counteract the IFN responses. Our showed Idarubicin (IDR) displays anti-EV71 activities in sub-cytotoxic concentrations. Moreover, IDR has recently been reported to increase IFN stimulating response element (ISRE) activity. However, the molecular mechanism of IDR underlying the IFN signal induction and antiviral effects remains to be explored. Materials and Methods: Western blot was used to probe the protein level of components in the IFN-activating pathway. The ISRE activity was measured by the dual luciferase assay. Confocal analysis was used to probe the localization of the interferon regulatory factor 9(IRF9). IFA and cytotoxicity assay were used to measure the combined effects of IDR and IFN-β. Results: We showed IDR promoting IFN signaling during EV71 infection. IDR raised the protein level of IRF9 but not in other components in the JAK-STAT pathway. IDR treatment resulted in IRF9 translocation from the cytoplasm to the nucleus. Finally, synergistic effect against EV71 infection was observed yet it also lowered down the cell viability. We have attempted to elaborate the molecular mechanisms underlying the type I IFN-activating pathway induced by IDR in the context of EV71 infection. This finding could lead to develop a broad-spectrum antiviral drug based on IFN signal stimulation.
