* 投稿摘要: Quercetin is a most typical flavonols that was found in vegetables and fruits. It had been reported to induce cytotoxic effects on many human cancer cells. In this study, we investigated the cytotoxic effects of Quercetin on human gastric carcinoma cell lines (AGS) in vitro. Treatments of AGS cells with Quercetin significantly decreased the percentage of viable cells through the induction of apoptosis and these effects are in concentration-dependent manner. To define the mechanism underlying the cytotoxic effects of Quercetin, we investigated the critical molecular events known to regulate the apoptotic machinery. We used flow cytometry analysis and the data showed that Quercetin promoted reactive oxygen species, NO and Ca2+ production, decreased the mitochondrial membrane potential (ΔΨm) and increased the activity of caspase-3, -8 and -9 in the AGS cells. Western blot analysis showed that Quercetin promoted the expression of Fas and FasL and increased the activity of caspase-3, -8 and -9, cytochrome c, Bax, apoptosis-inducing factor (AIF) and endonuclease G (Endo G) leading to apoptosis. Based on these observations, Quercetin induces apoptosis in AGS cells through Fas-, caspase- and mitochondrial-mediated pathways.
