* 投稿摘要: Human follicular growth directly affects oocyte quality and ovarian steroidogenesis. Follicular growth involves granulosa-thecal interactions encompassing endocrine, paracrine, and autocrine regulatory mechanisms, through which follicular size, follicular fluid volume and contents, and oocyte quality are determined. Eventually, fertilization rate, early embryogenesis, and the success rates of IVF-ET treatment cycles are affected. Hedgehog is a major regulatory gene during embryonic development. Previous reports indicated that Hedgehog and its downstream factors are expressed in murine granulosa cells and corpus lutea. Supplementation of Hedgehog protein in culture system can promote follicular growth and granulosa cell proliferation. Furthermore, it was shown that Hedgehog protein was secreted in the granulosa cell to promote thecal cell activities. Additionally, we have found in our previous studies that Hedgehog protein is enriched in porcine follicular fluid and oocyte maturation is promoted with Hedgehog supplementation under in vitro culture conditions. In contrast, Hedgehog signaling blockage by cyclopamine (a Hedgehog inhibitor) leads to insufficient oocyte maturation, suggesting that Hedgehog contents in follicular fluid may affect oocyte quality. However, whether Hedgehog protein is enriched in human follicular fluid and how does it affect human oocyte quality remain not yet reported. In this study, we investigated 119 oocytes undergoing IVF-ET treatment cycles from women with infertility. Follicular fluid Hedgehog protein contents were measured by using enzyme-linked immunological assay (ELISA) analysis and the data were correlated with follicular diameter, oocyte maturity, fertilization rate, pre-implantation embryogenesis, and implantation rate. The results showed that Hedgehog contents in the follicular fluid positively affect oocyte maturation and subsequent success rates of IVF-ET. The present results may be used for assessment of oocyte quality and may provide the baseline information for promoting IVF-ET success rate through Hedgehog supplementation.
