* 投稿摘要： Mortality and morbidity of cancer did not decline mainly resulting from tumor metastasis, especially hepatocellular carcinoma (HCC) in Taiwan. Resveratrol and its analogous have been verified to possess various biological benefits including antioxidative, anti-inflammatory, anti-cancer and antimetastatic in the previous studies. However, the detailed effects and molecular mechanisms of resveratrol on the metastasis tumors were unclear. Thus, HCC cells were subjected to a treatment with resveratrol and then analyzed for the effects of resveratrol on the migration and invasion. Modified Boyden chamber assays revealed that resveratrol treatment significantly inhibited cells migration and invasion capacities of HCC cells. Results of zymography and western blotting showed that activities and protein levels of urokinase-type plasminogen activator (u-PA) were inhibited by resveratrol. Testing of mRNA level, real-time PCR and promoter assays evaluated the inhibitory effects of resveratrol on u-PA expression in HCC cells. The chromatin immunoprecipitation (ChIP) assay for analyzing genomic DNA sequences bound to these proteins which showing reactive in transcription protein of nuclear factor SP1 that was inhibited by resveratrol. The expression of SP1 was measured by the Western blot analysis that revealed that the DNA-binding activity in nuclear decreasing by treatment of resveratrol. In conclusion, resveratrol inhibition of u-PA expression and metastasis of HCC cells as a powerful chemopreventive agent. The inhibitory effects are associated with downregulation of the transcription factors SP1 signaling pathways.
