* 投稿摘要： A 74-year-old man experienced a traffic accident and suffered from severe injury of the left leg with a soft tissue defect. Roentgenography revealed open comminuted fractures of the left tibia and fibula. Wound culture revealed yeast-like colonies on the agar. The results of API-20C (BioMerieux, France), urease test, and India ink staining indicated that this bacterium might be Cryptococcus albidus. Unexpectedly, PCR products did not support Cryptococcus albidus. Some sequences of the PCR products showed segments of Aureobasidium pullulans. To confirm this result, specific primers were designed for further identification and sequencing. The nucleotide sequences of the internal transcribed spacer (ITS) regions of rRNA genes (including ITS1, 5.8S rRNA gene, and ITS2) were analyzed with the BLAST program (NCBI). The sequences showed 99% (523/527) identity with Aureobasidium pullulans (accession number DQ680685.1). Analyzing the variable sequence area within fungal ribosomal RNA genes (ITS1 and ITS2) plays an important role in characterizing and identifying fungi. Although misidentification of bacteria occurs rarely, physicians and laboratorians should raise their suspicion of Aureobasidium pullulans when an atypical mucoid yeast isolate is found, especially if the colonies turn black after prolonged incubation even through a commercial identification system recornizes them as Cryptococcus spp.
