* 投稿摘要： BACKGROUND: The sensitivity in cytology diagnosis of malignant pleural effusion is insufficient nowadays due to the similarity of the reactive methothelial cells and malignant cells. VEGF-D was one of the key factors in tumor lymphangiogenesis and metastasis. Therefore, the aim of this study was to evaluate the value of VEGF-D in the aid of malignant pleural effusion diagnosis. METHODS: A total of 217 pleural effusions was collected from 2010 April to 2012 July. 103 samples including 81 malignant and 22 benign cases were made into the cell blocks for the immunocytochemical staining of VEGF-D expression. Another 114 samples (41 malignant and 73 benign cases) were subjected to ELISA for the protein level of VEGF-D. RESULTS: In total of 122 malignant pleural effusions included 69 lung adenocarcinoma (56.6 %), 19 breast infiltrating ductal carcinoma (15.5 %), 10 colorectal adenocarcinoma (8.2 %) and 24 others (19.7 %). Among the immunocytochemical stain, VEGF-D expression was significantly different between malignant (92.6 %) and benign cases (40.9 %) with p < 0.001 in either nuclear or cytoplasmic staining. In addition, 3 of 4 lung small cell carcinoma also showed positive results of VEGF-D expression. However, some lymphocytes also showed nuclear staining of VEGF-D. On the other hand, ELISA for the VEGF-D protein levels failed to demonstrate the difference between malignant and benign pleural effusionas. CONCLUSIONS: Malignant pleural effusion among various kinds of tumor origin, 92.6 % showed positive expression of VEGF-D. Immunocytochemical staining of VEDF-D might be a naval useful marker in aid of malignant pleural effusion diagnosis.
